用49个RAPD引物对烤烟13个抗赤星病品种和15个感赤星病品种基因组DNA进行RAPD分析,找到了一个和烤烟抗赤星病表型密切相关的DNA片段,此片段长760bp,命名为S220-760。通过对长勃黄和净叶黄的RAPD分析表明,S220-760为13个抗病品种所共有和15个感病品种没有。因此,S220-760可作为烤烟抗赤星病基因紧密连锁的RAPD标记。这为下一步钓取烤烟赤星病基因的工作打下了基础。
Forty-nine RAPD primers were used to analyze the genomic DNA of 13 resistant varieties and 15 susceptible varieties of Flue-cured tobacco by RAPD. A DNA fragment closely related to the resistance phenotype of Flue-cured tobacco was found. The fragment was 760bp and named S220 -760. RAPD analysis of Changbaihuang and Jingyehuang showed that S220-760 was shared by 13 resistant cultivars and 15 susceptible cultivars. Therefore, S220-760 can be used as a RAPD marker for the close linkage of Fusarium wilt resistance genes to brown spot disease. This laid the foundation for the next step of fishing for the brown spot disease gene work.
