目的寻找微囊藻毒素LR与宿主细胞(AGS)作用后的差异蛋白。方法利用双向电泳分离微囊藻毒素LR与胃黏膜AGS细胞相互作用前后的蛋白,应用ImageMaster 2Dv3.1软件对蛋白图谱进行比较,对目的蛋白进行基质辅助激光解析及电离飞行时间质谱分析,应用Mascot软件进行蛋白搜库。结果共监测到4个差异蛋白斑点,经质谱分析鉴定证明为两种蛋白,分别为热休克蛋白60与烷基过氧化物还原酶。结论微囊藻毒素LR与宿主细胞(AGS)作用后,宿主细胞热休克蛋白60与烷基过氧化物还原酶两种蛋白表达异常,其相关机制尚待进一步阐明。为进一步探讨蓝藻微囊藻毒素与宿主细胞作用后续致病过程提供了新的思路。
Objective To find the differential proteins after the action of microcystin LR and host cells (AGS). Methods Two-dimensional electrophoresis was used to separate proteins before and after interaction between microcystin-LR and gastric mucosa AGS cells. The protein profiles were compared with ImageMaster 2Dv3.1 software. Matrix-assisted laser and ionization time-of-flight mass spectrometry Software for protein search library. Results A total of four spots of differential proteins were detected and identified as two proteins by mass spectrometry analysis, namely, heat shock protein 60 and alkyl peroxiredoxin reductase. Conclusions The expression of heat shock protein 60 and alkylperoxide reductase in host cells is abnormal after the action of microcystin LR and host cells (AGS). The related mechanism remains to be elucidated. This provided a new idea for further research on the subsequent pathogenicity of cyanobacteria microcystins to host cells.
