目的协作标定肝素钠待标品的相对分子质量。方法高效凝胶色谱和多角度激光光散射仪联用(HGPC-MALLS)法,色谱柱和流动相等色谱条件各实验室自定,本实验采用的流速为0.5 m L·min-1,柱温为35℃,检测器为多角度激光光散射仪联用和示差折光检测器联用。结果本实验采用4种色谱条件,不同的进样体积,分别测定了待标品的相对分子质量,结果在报告值中间。结论不同实验室之间的结果存在较大差异,建议组织方进一步优化实验方法。
Objective To determine the relative molecular mass of heparin sodium to be labeled. Methods High-performance gel permeation chromatography (GPC) and multi-angle laser light scattering (HGPC-MALLS) were used in this study. The flow rate was 0.5 m L · min-1 and the column temperature 35 ℃, the detector is a multi-angle laser light scattering instrument combined with the refractive index detector. Results In this experiment, four kinds of chromatographic conditions and different injection volumes were used to determine the relative molecular mass of the products to be tested respectively. The results were reported in the middle. Conclusion The results of different laboratories are quite different, suggesting that the organizers further optimize the experimental method.
