目的观察庆大霉素对豚鼠耳蜗ACP的影响。方法以耳廓反射及酸性磷酸酶(ACP)为指标、观察正常及庆大霉素耳中毒豚鼠的耳蜗组织。结果正常豚鼠耳蜗内ACP主要集中在耳蜗的毛细胞内,并显示出毛细胞的排列形式;庆大霉素耳中毒的豚鼠耳蜗毛细胞内ACP出现位移、显色变淡或消失。耳廓反射阈值增加越明显者,ACP变化越显著。结论实验表明庆大霉素引起的耳蜗毛细胞的损坏与耳蜗毛细胞内溶酶体膜破裂释放出的ACP等水解酶的自溶性损坏有关。
Objective To observe the effect of gentamicin on cochlear ACP in guinea pigs. Methods Auricle reflex and acid phosphatase (ACP) were used as indices to observe the cochlea of normal and gentamicin ototoxicity guinea pigs. Results ACP in normal guinea pig cochleas was mainly concentrated in the hair cells of the cochlea and showed the arrangement of hair cells. The ACP in cochlear hair cells of gentamicin ototoxicity shifted and disappeared. The more obvious the increase of auricle reflex threshold, the more significant change of ACP. Conclusions Experiments show that gentamicin-induced cochlear hair cell damage and cochlear hair cell lysosomal membrane rupture release of ACP and other hydrolases autolysis damage.
