目的探讨氧化低密度脂蛋白(ox-LDL)及17β-雌二醇(E2)对体外培养的人脐静脉内皮细胞(HUVECs)血管紧张素转化酶II(ACE2)表达的影响。方法以不同浓度的ox-LDL(20,40,80mg/L)及E2(2,10,50nmol/L)分别单独刺激HUVECs 24h并以40mg/L的ox-LDL与不同浓度的E2(2,10,50nmol/L)共同刺激HUVECs 24h。逆转录聚合酶链反应(RT-PCR)检测ACE2 mRNA的表达情况。结果与对照组比较,ox-LDL单独刺激组ACE2 mRNA表达分别降低为0.74±0.08,0.52±0.07和0.31±0.06(均P<0.01),E2单独刺激组ACE2 mRNA表达分别增加为1.54±0.19,2.43±0.28和2.89±0.26(均P<0.01);40mg/L的ox-LDL与2,10,50nmol/LE2共同刺激组,ACE2 mRNA表达分别为40mg/Lox-LDL单独刺激组的1.95±0.38,2.56±0.32和3.22±0.37(3组间均P<0.05)。结论ox-LDL呈浓度依赖性下调ACE2 mRNA表达;E2呈浓度依赖性上调ACE2 mRNA表达,且E2呈正向量效关系抑制ox-LDL引起的ACE2表达下调。
Objective To investigate the effects of ox-LDL and 17β-estradiol on the expression of angiotensin-converting enzyme II (ACE2) in cultured human umbilical vein endothelial cells (HUVECs). Methods HUVECs were stimulated with different concentrations of ox-LDL (20, 40, 80mg / L) and E2 (2,10,50nmol / L) 10, 50nmol / L) co-stimulated HUVECs 24h. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of ACE2 mRNA. Results Compared with the control group, the expression of ACE2 mRNA in ox-LDL alone group was decreased to 0.74 ± 0.08, 0.52 ± 0.07 and 0.31 ± 0.06, respectively (all P <0.01), and the ACE2 mRNA expression in E2 alone group increased to 1.54 ± 0.19, 2.43 ± 0.28 and 2.89 ± 0.26 respectively (all P <0.01). Compared with 40 mg / L ox-LDL and 2,10,50 nmol / LE2 co-stimulation group, ACE2 mRNA expression was 1.95 ± 0.38 , 2.56 ± 0.32 and 3.22 ± 0.37 (all P <0.05). Conclusion Ox-LDL down-regulates the expression of ACE2 mRNA in a concentration-dependent manner. E2 up-regulates the expression of ACE2 mRNA in a concentration-dependent manner. E2 has a positive dose-effect relationship and inhibits the down-regulation of ACE2 expression induced by ox-LDL.
