应用荧光探针和激光共聚焦扫描技术观察高糖、高胰岛素、高胰岛素原、肿瘤坏死因子及佛波酯类物质等及糖尿病因素对培养的人脐静脉内皮细胞胞内游离钙离子的作用。高糖、高胰岛素原及肿瘤坏死因子(1~3.5Mu/L)可引起群体细胞内钙离子浓度明显升高(P<0.01),而0.2Mu/L肿瘤坏死因子作用较弱(P<0.05),高胰岛素作用不明显(P>0.05)。动态观察可见,高糖、高胰岛素原、肿瘤坏死因子和佛波酯类物质可引起单个内皮细胞胞内钙离子浓度升高,以核区为显著。肿瘤坏死因子和高胰岛素可诱发“钙振荡”,大剂量佛波酯类物质促使细胞排钙。提示在糖尿病血管并发症中许多致病因子可能通过影响内皮细胞胞内、外钙离子水平而导致其一系列功能和结构损害。
The effects of high glucose, high insulin, high proinsulin, tumor necrosis factor, phorbol ester and other diabetes factors on intracellular free calcium in cultured human umbilical vein endothelial cells were observed by fluorescence probe and confocal laser scanning microscopy. High glucose, high proinsulin and tumor necrosis factor (1 ~ 3.5Mu / L) caused a significant increase in intracellular calcium concentration (P <0.01), while 0.2Mu / L tumor necrosis factor (P <0.05), and the effect of hyperinsulin was insignificant (P> 0.05). Dynamic observation shows that high glucose, high proinsulin, tumor necrosis factor and phorbol esters can cause a single endothelial cell intracellular calcium concentration increased to the nuclear region as a significant. Tumor necrosis factor and high insulin can induce “calcium oscillation”, high doses of phorbol ester substances to promote cell row calcium. It is suggested that many virulence factors in diabetic vascular complications may cause a series of functional and structural damage by affecting the intracellular and extracellular calcium levels of endothelial cells.
