目的通过心肌酶和热休克蛋白(HSP)70的表达来观察慢性应激反应中丹红注射液对大鼠心肌的保护作用。方法将健康成年SD大鼠55只随机分为空白对照组、丹红对照组、慢性应激组和慢性应激丹红组。空白对照组不进行慢性应激处理,也不注射丹红注射液;丹红对照组不进行慢性应激处理,仅每日经腹腔注射丹红注射液10 ml/kg;慢性应激组按照慢性应激反应模型进行处理;慢性应激丹红组在按照慢性应激反应模型进行处理的同时每日经腹腔注射丹红注射液10 ml/kg。待慢性应激持续到设定时间时(3周),剖杀4组大鼠,(1)观察各组大鼠心肌酶的变化。(2)同时采用免疫组织化学法检测各组大鼠心肌组织中HSP70的表达情况。结果 (1)与空白对照组比较,丹红对照组、慢性应激组和慢性应激丹红组大鼠乳酸脱氢酶(LDH)和肌酸激酶同工酶(CK-MB)显著升高(P<0.01)。慢性应激组LDH和CK-MB较丹红对照组显著升高(P<0.05),慢性应激丹红组LDH和CK-MB较慢性应激组显著降低(P<0.05);而丹红对照组和慢性应激丹红组之间无显著性差异。(2)丹红对照组、慢性应激组和慢性应激丹红组心肌组织HSP70表达均强于空白对照组。丹红对照组和慢性应激组之间HSP70表达无显著性差异。慢性应激丹红组心肌组织HSP70表达强于丹红对照组或慢性应激组。结论丹红注射液的使用可有效抑制慢性应激大鼠LDH、CK-MB的升高,还可增高慢性应激大鼠心肌组织中HSP70的表达。
Objective To observe the protective effect of Danhong injection on myocardium in chronic stress induced by the expression of myocardial enzymes and heat shock protein (HSP) 70. LDH and CK-MB in chronic stress group were significantly higher than those in control group (P <0.05), but LDH and CK-MB in chronic stress Danhong group were significantly lower than those in chronic stress group (P <0.05) There was no significant difference between the control group and the chronic stress Danhong group. (2) The expression of HSP70 in Danhong control group, chronic stress group and chronic stress Danhong group were stronger than those in blank control group. There was no significant difference in HSP70 expression between Danhong control group and chronic stress group. The expression of HSP70 in chronic stress Danhong group was stronger than that in Danhong control group or chronic stress group. Conclusion Danhong injection can effectively inhibit the increase of LDH and CK-MB in chronic stress rats and increase the expression of HSP70 in chronic stress rats.
