目的:观察活血化瘀中药对C6脑肿瘤干细胞(BTSCs)侵袭能力及组织型金属蛋白酶抑制因子1(TIMP1)mRNA表达的影响。方法:将20只大鼠分为对照组(生理盐水)、B组(桃红四物汤)、C组(血府逐瘀汤)、D组(抵当汤)各5只,给予相应药物灌胃10天后,分离血清分别与C6 BTSCs体外共同培养,进行Boyden体外细胞侵袭试验检测C6 BTSCs侵袭能力变化,荧光定量PCR检测C6 BTCs的TIMP1 mRNA表达变化。结果:B组、C组、D组的C6 BTSCs细胞侵袭率降低、TIMP1mRNA表达升高,与对照组比较,差异均有显著性意义(P<0.05)。结论:活血化瘀中药能抑制C6 BTSCs的侵袭能力,其机理可能在于促进TIMP1 mRNA表达,增强对基质金属蛋白酶的抑制作用。
Objective: To observe the effect of traditional Chinese medicine of promoting blood circulation and removing blood stasis on invasiveness of rat brain tumor stem cells (BTSCs) and expression of tissue inhibitor of metalloproteinase 1 (TIMP1). Methods: Twenty rats were divided into control group (normal saline), B group (Taohongsiwutang), C group (Xuefuzhuyu Decoction) and D group (Dangdang Decoction) After 10 days, the sera were separated and co-cultured with C6 BTSCs in vitro. The invasion of C6 BTSCs was detected by Boyden cell invasion assay. The expression of TIMP1 mRNA in C6 BTCs was detected by real-time PCR. Results: The invasion rates of C6 BTSCs in groups B, C and D were decreased and the expression of TIMP1 mRNA was increased. Compared with the control group, the differences were significant (P <0.05). CONCLUSION: Chinese Herbs for Activating Blood Circulation and Removing Blood Stasis can inhibit the invasion ability of C6 BTSCs. Its mechanism may lie in promoting the expression of TIMP1 mRNA and enhancing the inhibition of matrix metalloproteinase.
