采用人胎脑垂体前叶分散细胞建立了无血清体外培养模型。大量垂体细胞对 GH、PRL、TSH 等六种垂体前叶激素 PAP 免疫细胞化学标记呈阳性反应。应用放免方法测定了不同时间培养液中 GH 等激素含量。结果表明分散垂体细胞在无血清培养液中能够存活,并具有合成和分泌激素的功能。无血清培养液使垂体细胞脱离了机体复杂的调控机制和培养基中血清成分的影响,能够人为地控制条件,为内分泌学、神经内分泌学的深入研究提供了理想的实验模型。
A serum-free in vitro culture model was established using human fetal brain anterior pituitary dispersed cells. A large number of pituitary cells of GH, PRL, TSH and other six kinds of pituitary prolactin PAP immunocytochemical staining was positive. Radionuclide method was used to determine the content of GH and other hormones in culture medium at different time. The results show that dispersed pituitary cells can survive in serum-free medium and have the function of synthesizing and secreting hormones. Serum-free culture fluid pituitary cells from the body’s complex regulatory mechanisms and serum composition of the media, can artificially control the conditions for endocrinology, neuroendocrine studies provide an ideal experimental model.
