目的探讨胃癌组织中hTERT、hMSH2基因在胃癌发生中的作用和Hp致癌的可能机制。方法采用W-S法检测93例胃癌患者癌组织及癌旁组织中Hp感染情况;利用免疫组织化学方法检测93例胃癌患者癌组织及癌旁组织中hTERT、hMSH2的表达情况。结果(1)胃癌组织hTERT阳性率为79.6%,明显高于癌旁组织22.6%(P<0.01);Hp感染组hTERT蛋白表达阳性率高于非感染组(P<0.01);(2)胃癌组织hMSH2阳性率为65.6%,明显高于癌旁组织38.7%(P<0.01);Hp感染组hMSH2基因蛋白表达阳性率低于非感染组(P<0.05)。结论Hp诱导的端粒酶的表达增高和错配修复基因hMSH2的表达异常可能与胃癌的发生有关。
Objective To investigate the role of hTERT and hMSH2 gene in the development of gastric cancer and the possible mechanism of Hp carcinogenesis. Methods The H-infection of cancer tissues and paracancerous tissues in 93 cases of gastric cancer was detected by W-S method. The expression of hTERT and hMSH2 in cancer tissues and paracancerous tissues of 93 patients with gastric cancer were detected by immunohistochemistry. Results (1) The positive rate of hTERT in gastric cancer was 79.6%, which was significantly higher than that in paratumor tissue (22.6%, P<0.01). The positive rate of hTERT protein expression in Hp infection group was higher than that in non-infected group (P<0.01); The positive rate of hMSH2 in tissue was 65.6%, which was significantly higher than that in para-cancer tissue (38.7%) (P<0.01). The positive rate of hMSH2 gene protein expression in Hp-infected group was lower than that in non-infected group (P<0.05). Conclusion The increased expression of telomerase induced by Hp and the abnormal expression of mismatch repair gene hMSH2 may be related to the occurrence of gastric cancer.
