为揭示初筛所得7株活树附生铁皮石斛内生真菌对宿主生长及代谢成分的影响,该研究观察测量了接菌与未接菌铁皮石斛组培苗形态特征与农艺性状,采用苯酚-硫酸法与热浸法测定组培苗茎总多糖含量与浸出物含量,柱前衍生HPLC分析茎多糖中单糖组成,HPLC分析茎醇溶性成分。研究表明,供试7个菌株可改变铁皮石斛组培苗茎节变紫进程,并影响茎糖类及醇溶性成分组成及含量积累,除DO34外均能促进组培苗生长与提高多糖含量,除DO12外均能促进甘露糖含量积累,DO18,DO19,DO120可促进醇溶性浸出物含量积累,综合生长及代谢成分多项指标筛选出DO14,DO18,DO19与DO120 4个优良菌株,为铁皮石斛内生真菌和宿主相互作用机制研究与微生物工程研究提供了菌种材料与研究基础。
In order to reveal the effects of endophytic fungi on the growth and metabolism of seven Dendrobium candidum samples from primary screening, the morphological and agronomic characters of the tissue culture seedlings of inoculated and non-inoculated Dendrobium candidum were observed. Phenol- Sulfate method and hot-dipping method were used to determine the total polysaccharides content and the content of the extract in the stem of the tissue culture. The pre-column derivatization HPLC was used to analyze the monosaccharide composition in the stem polysaccharides. The results showed that the seven strains tested could change the process of purple process of stems and leaves of D. officinale and affected the composition and content accumulation of stems and sugars and alcohol-soluble components. All of them could promote the growth of tissue culture seedlings and increase the content of polysaccharides except DO34, DO18, DO19 and DO120 could promote the accumulation of alcohol-soluble extract, and the four excellent strains of DO14, DO18, DO19 and DO120 were screened out by comprehensive index of growth and metabolism, The mechanism of endophytic fungi and host interaction and microbiological engineering provide the material of bacteria and research basis.
