目的观察启膈散对4NQO诱导食管癌小鼠生存质量、体重、脏器重量和指数、食管组织病理形态、细胞因子及淋巴细胞亚群的干预作用。方法 0.1mg·ml~(-1)4NQO水诱导小鼠食管癌模型,16周后撤除干预因素,观察进食及饮水并称重,32周后处死称重脏器并计算脏器指数,观察食管组织,检测外周血中IL-10、TNF、IFN-γ、MCP-1、IL-6、IL-12p70等细胞因子,检测外周血中淋巴细胞亚群(CD3~+/CD19~+,CD4~+/CD8~+)。结果启膈散组小鼠形体、活动度正常,体重接近正常组;脾、肝重量高于模型组(P<0.05),肾、心、肺、肝指数降低(P<0.05);镜下观食管基底细胞层有轻度增生,角质层液化基本消失,其它均正常;与模型组比可上调IL-10细胞因子(P<0.05),降低CD3~+、CD19~+、CD3~+/CD19~+、CD8~+,提高CD4~+/CD8~+(P<0.05)。结论启膈散能改善生存质量、保护脏器、保护组织病理损害;通过提高IL-10细胞因子水平提升免疫功能,提高CD3~+、CD19~+、CD8~+淋巴细胞,纠正CD4~+/CD8~+比例降低而改善免疫功能失衡及免疫抑制状态。
Objective To observe the effects of Qige San on 4NQO-induced mouse esophageal cancer quality of life, body weight, organ weight and index, esophageal histopathology, cytokines and lymphocyte subsets. Methods The mice model of esophageal cancer was induced by 0.1 mg · ml ~ (-1) 4NQO water. After 16 weeks, the intervention factors were removed, the food intake and drinking water were taken and weighed. After 32 weeks, the weighed organs were sacrificed and the organ index was calculated. The levels of IL-10, TNF-α, MCP-1, IL-6 and IL-12p70 in peripheral blood were measured and the levels of CD3 ~ + / CD19 ~ + / CD8 ~ +). Results The body weight, body weight and body weight of QGG group were normal, and the weights of spleen and liver were higher than those of model group (P <0.05). The indexes of kidney, heart, lung and liver were decreased (P <0.05) The esophageal basal cell layer had slight hyperplasia, and the keratinized layer almost disappeared, all other were normal. Compared with the model group, IL-10 cytokine (P <0.05) and CD3 +, CD19 +, CD3 + / CD19 ~ +, CD8 ~ + increased CD4 ~ + / CD8 ~ + (P <0.05). Conclusions Qige Powder can improve the quality of life, protect the organs and protect the histopathological damage. It can improve the immune function, increase the CD3 +, CD19 +, CD8 + lymphocytes and the CD4 + / CD8 + lymphocytes by increasing the level of IL-10. CD8 ~ + ratio decreased to improve immune function imbalance and immunosuppressive status.
