应用一期法测定了16~22周胎儿肝、脾、肾组织中凝血因子VIII:C(FVIII:C)活性,发现肾组织中FVIII:C活性为1.723IU/ml,高于肝和脾(P<0.01)。三种组织细胞培养液中肾细胞培养液的FVIII:C活性始终较高。LSAB法免疫组化检测发现在肾远曲小管和集合管上皮细胞以及肝的胆管上皮细胞和肝、脾血管内皮细胞FVIII:CAg呈阳性。逆转录-多聚酶链反应证实三种组织中均有FVIIImRNA表达。表明在16~22周胎龄期肝、脾、肾均可合成FVIII:C,其中以肾最为明显。
The FVIII: C (FVIII: C) activity in fetal liver, spleen and kidney at 16-22 weeks was measured by the first-phase method. The FVIII: C activity in renal tissue was found to be 1.723 IU / ml higher than that of the liver and spleen (P <0.01). FVIII: C activity of renal cell culture fluid in all three tissue cell cultures was consistently high. LSAB immunohistochemical detection found in renal distal convoluted tubules and collecting duct epithelial cells and liver bile duct epithelial cells and liver and spleen vascular endothelial cells FVIII: CAg positive. Reverse transcription-polymerase chain reaction confirmed FVIII mRNA expression in all three tissues. The results showed that FVIII: C could be synthesized in the liver, spleen and kidney in the gestational age of 16-22 weeks, of which kidney was the most obvious.
