目的探讨HSV 2在体内诱导细胞凋亡的效果、形态学特点、NF κB蛋白的表达和TNF的促进凋亡作用。方法用HSV 2 333株感染小鼠阴道 ,光镜下进行原位观察 ,TUNEL末端标记染色显示凋亡的细胞 ,免疫组化技术检查NF κB蛋白的表达。结果感染后的第 1天粘膜上皮内即出现大量的凋亡细胞 ,第 3- 6天凋亡细胞的数量及上皮内的分布范围达最高水平。凋亡细胞被TUNEL标记染成棕褐色 ,经免疫组化技术证实凋亡细胞有NF -κB蛋白的表达。结论HSV 2 333株阴道感染可同时诱导细胞坏死及凋亡 ,NF κB蛋白的表达参与了细胞凋亡的发生 ,TNF可以促进HSV 2在体内诱导的细胞凋亡。细胞凋亡可能在限制病毒产生子代及限制感染区域扩展中起重要作用。
Objective To investigate the effect of HSV 2 on inducing apoptosis in vivo, the morphological features, the expression of NF κB and the promotion of apoptosis induced by TNF. Methods The vagina was infected with HSV 2 333 and observed in situ by light microscopy. Apoptotic cells were stained with terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL). The expression of NF-κB protein was detected by immunohistochemistry. Results A large number of apoptotic cells were found in the mucosal epithelium on the first day after infection. The number of apoptotic cells and the distribution of intraepithelial epithelial cells on day 3-6 reached the highest level. TUNEL-stained apoptotic cells were stained tan, and the expression of NF-κB protein in apoptotic cells was confirmed by immunohistochemistry. Conclusions Vaginal infection with HSV 2 333 can induce necrosis and apoptosis at the same time. The expression of NF - κB is involved in the apoptosis of HSV 2 333 strain. TNF can promote the apoptosis induced by HSV 2 in vivo. Apoptosis may play an important role in limiting the generation of the virus and in limiting the expansion of the infected area.
