目的:研究抗纤I号对肝星状细胞(HSC)的影响及抗肝纤维化的机制.方法:用RT-PCR方法检测转化生长因子β1(TGF-β1)和血小板衍生生长因子(PDGF)mRNA在HSC的表达,用W estern B lot检测TGF-β1和PDGF蛋白的表达.同时,应用MTT法观察HSC的生长曲线.结果:抗纤I号复方药物血清作用于HSC后,与对照组和秋水仙碱组相比HSC增殖明显被抑制(P<0.05),同时TGF-β1和PDGF蛋白及mRNA的表达明显下降(P<0.05).结论:抗纤I号复方药物血清能够抑制HSC增殖,下调TGF-β1和PDGF蛋白及mRNA的表达,这可能是该药抗肝纤维化的分子机制之一.
Objective: To study the effect of anti-fibrosis I on hepatic stellate cells (HSC) and the mechanism of anti-hepatic fibrosis. Methods: Detection of transforming growth factor β1 (TGF-β1) and platelet-derived growth factor (PDGF) by RT-PCR Expression of mRNA in HSCs. The expression of TGF-β1 and PDGF protein was detected with W estern B lot. At the same time, the growth curve of HSC was observed by MTT assay. Results: After anti-fibrosis compound I serum acted on HSC, it was compared with the control group. Compared with colchicine group, proliferation of HSCs was significantly inhibited (P<0.05), while TGF-β1 and PDGF protein and mRNA expressions were significantly decreased (P<0.05). Conclusion: Anti-fibrosis I compound serum can inhibit the proliferation of HSCs. Down-regulation of TGF-β1 and PDGF protein and mRNA expression, which may be one of the molecular mechanisms of the anti-fibrosis of the drug.
