目的了解杭州市急性呼吸道感染儿童中WU多瘤病毒(WU polyomavirus,WUPy V)流行特征和基因型别。方法收集2011年3月至2013年6月间至浙江省儿童医院就诊的急性呼吸道感染患儿的鼻咽吸取物,用聚合酶链反应(PCR)法扩增VP2编码基因和VP2/1中间序列,测序后进行分型。选取代表性病毒株进行基因组测序。结果收集的722份标本中,10份为WUPy V阳性,检出率为1.4%。WUPy V感染主要发生在夏季、秋季和冬季。患儿平均年龄为13.6月龄,男女性别比为9∶1,性别差异无统计学意义(P=0.09)。WUPy V感染所致疾病为肺炎、喉炎和细支气管炎。WUPy V基因型别为Ⅰa型。代表性毒株HZ13024的基因组大小为5229 bp,与Gen Bank中WUPy V基因组序列相似性为99.8%~99.9%。结论杭州市儿童急性呼吸系统疾病中存在WUPy V感染,感染率较我国其他地区低,基因型别为Ⅰa型。
Objective To investigate the prevalence and genotype of WU polyomavirus (WUPy V) in children with acute respiratory infection in Hangzhou. Methods Nasopharyngeal aspirates of children with acute respiratory infection were collected from March 2011 to June 2013 in Zhejiang Children’s Hospital. The VP2 encoding gene and VP2 / 1 intermediate sequence were amplified by polymerase chain reaction (PCR) , Sequencing after sequencing. Select representative strains for genome sequencing. Results Of the 722 specimens collected, 10 were WUPy V positive with a detection rate of 1.4%. WUPy V infections occur mainly in summer, autumn and winter. The mean age of children was 13.6 months, with a sex ratio of 9: 1. There was no significant difference in gender (P = 0.09). The diseases caused by WUPy V infection are pneumonia, laryngitis and bronchiolitis. WUPy V genotype is Ⅰa. The representative strain HZ13024 has a genome size of 5229 bp, which is 99.8% -99.9% similar to the GenBank WUPy V genome sequence. Conclusions There is WUPy V infection in children with acute respiratory diseases in Hangzhou, the infection rate is lower than other parts of China and the genotype is Ⅰa.
