紫杉醇是治疗乳腺癌的抗微管解聚药。本文探讨紫杉醇对巨噬细胞分泌IL-6和TNF-α的影响,作为紫杉醇抗癌效应的另一机制。方法:正常鼠和乳腺癌瘤鼠腹腔巨噬细胞(PEM)与不同浓度的紫杉醇和大肠杆菌脂多糖(LPS)分别或混合培养。在24h和48h用ELISA及Western blot分析PEM分泌IL-6和TNF-α的变化。结果:用紫杉醇处理瘤鼠PEM24h,IL-6水平明显降低,与正常鼠PEM比较有非常显著性差异(P<0.01)。紫杉醇对PEM分泌IL-6的抑制作用可以被LPS拮抗。紫杉醇和LPS均能增强正常和瘤鼠PEM分泌TNF-α,两组间TNF-α水平有非常显著性差异(P<0.001)。Western blot表明紫杉醇能促进瘤鼠PEM的TNF-α合成。结论:紫杉醇促进瘤鼠PEM的TNF-α分泌和抑制IL-6的产生,佐以提高紫杉醇的杀瘤效应。
Paclitaxel is an anti-microtubule depolymerization drug for the treatment of breast cancer. This article explored the effect of paclitaxel on the secretion of IL-6 and TNF-α by macrophages as another mechanism for the anti-cancer effect of paclitaxel. METHODS: Peritoneal macrophages (PEM) from normal mice and breast cancer mice were incubated with different concentrations of paclitaxel and E. coli lipopolysaccharide (LPS) separately or mixed. The changes of IL-6 and TNF-α secreted by PEM were analyzed by ELISA and Western blot at 24h and 48h. RESULTS: After treated with paclitaxel for 24 hours, the IL-6 level of PEM was significantly lower than that of PEM in normal rats (P<0.01). The inhibitory effect of paclitaxel on the secretion of IL-6 by PEM can be antagonized by LPS. Both paclitaxel and LPS increased the secretion of TNF-α by normal and tumoral PEM. There was a significant difference in TNF-α levels between the two groups (P<0.001). Western blot showed that paclitaxel can promote the synthesis of tumor necrosis factor-α in PEM. Conclusion: Paclitaxel can promote the secretion of TNF-α and inhibit the production of IL-6 in PEM of tumor mice, and it can improve the tumoricidal effect of paclitaxel.
