为测定及分析甘蔗在不同收获时间蔗糖含量的变化,采用水处理甘蔗样品,采取高效液相色谱法,经Waters Sugar PakⅠ柱(6.5 mm×300 mm,10.0μm),Waters 2414示差折光检测器(RID)检测,流动相为高纯水,流速:1.0 mL·min-1,柱温:90℃,检测器温度:35℃.结果显示该法相对标准偏差(RSD)(n=6)<2%,回收率为95%~105%,蔗糖质量浓度为10~50 mg·mL-1.且甘蔗在不同的收割时间蔗糖含量变化较明显,基本在14:00~15:00时达到最大值.实验表明HPLC法能够较快且较准确测出甘蔗中蔗糖的含量.通过比较可知,甘蔗蔗茎中蔗糖含量是随着光合作用的强弱而变化的.
In order to measure and analyze the change of sucrose content of sugarcane at different harvest time, sugarcane samples were treated with water and analyzed by Waters Sugar PakⅠcolumn (6.5 mm × 300 mm, 10.0 μm), Waters 2414 refractive index detector RID), the mobile phase was high purity water, the flow rate was 1.0 mL · min-1, the column temperature was 90 ℃ and the detector temperature was 35 ℃ .The results showed that the relative standard deviation (RSD) was less than 2% The recoveries were 95% ~ 105%, sucrose concentration was 10 ~ 50 mg · mL-1, and sucrose content of sugarcane changed significantly at different harvesting time, reaching the maximum at 14: 00-15: 00. The results showed that the HPLC method could determine the content of sucrose in sugarcane quickly and accurately.Comparison shows that the content of sucrose in sugarcane cane changes with the intensity of photosynthesis.
