目的探讨奥沙利铂联合热疗对胃癌细胞系和实体瘤的体外疗效及其临床指导意义。方法采用ATP生物荧光法分析比较化疗药奥沙利铂(L-OHP)、顺铂(DDP)、氟尿嘧啶(5-Fu)、阿霉素(ADM)、丝裂霉素(MMC)分别与热疗联合对人胃癌细胞BGC-823的抑制率的差异;并采用体外HDRA法对比研究上述五种化疗药物联合热疗对48份胃癌组织的敏感性。结果五种化疗药在37℃~45℃温度区间内对BGC-823细胞的杀伤效应随温度的升高及作用时间的延长而增强,具有温度和时间依赖效应,其中L-OHP的热杀伤效应最强。43℃条件下作用60 min,不同化疗药对胃癌组织的敏感性为L-OHP>DDP>ADM>MMC>5-Fu。结论 L-OHP联合热疗对胃癌具有较强的协同杀伤效应,有可能是治疗胃癌理想的热化疗药物之一。
Objective To investigate the curative effect of oxaliplatin plus hyperthermia on gastric cancer cell lines and solid tumors in vitro and its clinical significance. Methods ATP biofluorescence assay was used to compare the chemotherapeutic drugs oxaliplatin (L-OHP), cisplatin (DDP), fluorouracil (5-Fu), doxorubicin (ADM) and mitomycin (MMC) The results showed that the sensitivity of the five chemotherapeutic agents combined with hyperthermia to 48 gastric cancer tissues was tested by in vitro HDRA. Results The killing effect of five chemotherapeutic drugs on BGC-823 cells at 37 ℃ ~ 45 ℃ increased with the increase of temperature and time, and had a temperature and time-dependent effect. The killing effect of L-OHP The strongest 43 ℃ for 60 min. The sensitivity of different chemotherapeutic agents to gastric cancer was L-OHP> DDP> ADM> MMC> 5-Fu. Conclusions L-OHP combined with hyperthermia has a strong synergistic killing effect on gastric cancer and may be one of the ideal thermochemotherapy drugs for gastric cancer.
