目的 探讨T细胞亚群及共刺激分子表达与系统性红斑狼疮免疫病理的关系。方法 采用免疫荧光标记技术和流式细胞仪检测技术对系统性红斑狼疮 (SLE)患者和正常人外周血T细胞亚群及外周血单个核细胞膜表面共刺激分子的表达进行分析。结果 ①与正常人相比 ,SLE患者CD3+ 、CD4+ T细胞亚群显著减少 ,CD4/CD8比例失调 ,双标记CD4+ CD2 8+ 、CD8+ CD2 8+ T细胞亦显著减少 ;②患者共刺激分子CD2 8表达显著降低 ,4-1BB分子表达显著增高 ;③患者T细胞CD4/CD8与抗核抗体呈负相关 ,共刺激分子 4- 1BB与抗核抗体、尿蛋白呈正相关 ,而与CD4+ CD2 8+ T细胞亚群呈负相关。结论 T细胞亚群及共刺激分子表达异常在系统性红斑狼疮发病机制中可能具有重要作用。
Objective To investigate the relationship between T cell subsets and costimulatory molecules expression and immunopathology in patients with systemic lupus erythematosus. Methods Immunofluorescence staining and flow cytometry were used to analyze the expression of costimulatory molecules in the peripheral blood T cell subsets and peripheral blood mononuclear cells in patients with systemic lupus erythematosus (SLE) and normal controls. Results ① Compared with normal controls, the percentage of CD3 + and CD4 + T cells in SLE patients was significantly decreased and the proportion of CD4 / CD8 was downregulated. The numbers of double labeled CD4 + CD28 + and CD8 + CD28 + T cells were also significantly decreased. The expression of 4-1BB was significantly increased; ③The CD4 / CD8 of T cells was negatively correlated with antinuclear antibody, and the co-stimulatory molecule 4-1BB was positively correlated with antinuclear antibody and urine protein, but not with CD4 + CD2 + T Cell subpopulation was negatively correlated. Conclusion The abnormal expression of T cell subsets and costimulatory molecules may play an important role in the pathogenesis of systemic lupus erythematosus.
