目的:通过对自身免疫性溶血性贫血(autoimmune hemolytic anemia,AIHA)患者抗体生成细胞(antibodysecreting cell,ASC)的检测,分析Sysmex XE2100分析仪与流式细胞仪的相关性及散点图的分布特征。方法:检测65例AIHA患者外周血白细胞并绘制SSC-SFL通道散点图;BD FACScalibur流式细胞仪检测ASC;显微镜分类计数200个白细胞中的ASC作为最终判定依据。结果:>9个ASC/200WBC,流式细胞仪检测>350个/μl,XE2100为级分布;1~6个ASC/200WBC,流式细胞仪检测100~300个/μl,XE2100为级分布;而在XE2100为级分布时,显微镜检查为阴性,流式细胞仪检测50个/μl左右。结论:检测ASC,流式细胞仪结果为最佳,而XE2100虽然不能准确定量,但其高速高可信度的分布图像是临床应用的重要补充。
OBJECTIVE: To analyze the correlation between the Sysmex XE2100 analyzer and flow cytometry and the distribution features of scattergram by detecting the antibody-producing cells (ASC) of patients with autoimmune hemolytic anemia (AIHA) . Methods: Peripheral blood leukocytes were detected in 65 patients with AIHA and SSC-SFL channel scatter plot was drawn. The ASC was detected by BD FACScalibur flow cytometry. The ASC was classified by microscope in 200 leukocytes as the final judgment. Results:> 9 ASC / 200WBC, flow cytometry> 350 cells / μl, XE2100 was level distribution; 1 ~ 6 ASC / 200WBC, flow cytometry 100 ~ 300 cells / In the XE2100 level distribution, microscopy was negative, flow cytometry 50 / μl or so. Conclusion: The results of detection of ASC and flow cytometry are the best. Although XE2100 can not be accurately quantified, its high-speed and high-confidence distribution image is an important complement to clinical application.
