目的:优选卷柏抗SMMC-7721肝肿瘤组分总黄酮的提取工艺。方法:选取提取时间、提取次数、乙醇用量及体积分数为考察因素,以人肝癌细胞SMMC-7721抑制率为指标的正交试验结果优选总黄酮和穗花杉双黄酮提取量综合评分的权重系数比,并通过SPSS 17.0软件对有效组分与药效学指标进行Pearson相关系数分析。采用HPLC测定穗花杉双黄酮含量,流动相乙腈-0.5%乙酸水(33∶67),检测波长269 nm。结果:最佳提取工艺条件为加10倍量70%乙醇回流提取1次,每次3 h;穗花杉双黄酮含量与总黄酮含量的权重系数分别为0.7,0.3,反映含量指标与抑制率呈一定相关性。结论:以有效成分含量代表药效作为正交试验指标是科学合理的,优选的提取工艺稳定可行,为中药有效成分的提取提供新思路。
