人体骨髓细胞长期培养可维持造血活性达2—3个月。10例人体骨髓有核细胞植入一种复合液体培养体系,观察到一层融合的粘壁细胞层,包含有骨髓造血细胞和各种骨髓基质细胞。在粘壁细胞层上方为非粘壁细胞层,其细胞来自粘壁层。动态观察粘壁层和非粘壁层的生长情况,可发现细胞的生长和组成有一定规律性变化。定期测定非粘壁细胞层的CFUGM产量,可推断培养体系中粒系祖细胞的生长情况。
Long-term human bone marrow cell culture can maintain hematopoietic activity of 2-3 months. Ten human bone marrow nucleated cells were implanted into a complex liquid culture system and a layer of fused muco-cellular layers including bone marrow hematopoietic cells and various bone marrow stromal cells was observed. Above the parietal cell layer is a layer of non-adherent parietal cells from which the cells are derived. Dynamic observation of the growth of viscous and non-viscous layer growth, we can find the cell growth and composition of a certain regularity. Regular measurement of CFUGM production in non-adherent cell layers can infer the growth of myeloid progenitor cells in the culture system.
