目的:研究银杏叶提取物(Ginkgo Biloba extract,GBE)对大鼠肾缺血再灌注和顺氯氨铂(顺铂)所致肾近曲小管转运功能损伤的保护作用。方法:采用体外肾皮质薄片孵育法,以肾薄片蓄积对氨基马尿酸(p-aminohippurate,PAH)的量间接反映肾脏近曲小管的转运功能。结果:GBE能促进大鼠肾皮质薄片摄取PAH,并能抵抗肾缺血再灌注和顺铂所致的大鼠肾皮质薄片摄取PAH的减少。此外,GBE同时减少了肾皮质中ATP的浓度,而增加了K~+浓度。ATP的减少可能与GBE增加Na~+-K~+-ATP酶活性而消耗了ATP有关,因为PAH的转运是耗能的过程。但这一结论还需进一步研究。结论:GBE促进肾薄片蓄积PAH的作用可以抵消大鼠急性肾衰竭时所致的PAH蓄积的减少。
Objective: To study the protective effect of ginkgo biloba extract (GBE) on renal proximal tubular injury induced by renal ischemia / reperfusion and cisplatin (cisplatin) in rats. Methods: The in vitro renal cortical slice incubation method was used to indirectly reflect the transport function of renal proximal tubule with the amount of p-aminohippurate (PAH) accumulated in the renal lamina. RESULTS: GBE promoted the uptake of PAH by rat renal cortical slices and the decrease of PAH uptake induced by renal ischemia-reperfusion and cisplatin-induced rat renal cortical slices. In addition, GBE reduced the concentration of ATP in renal cortex and increased the concentration of K ~ +. The decrease of ATP may be related to the increase of Na ~ + -K ~ + -ATPase activity by GBE, because the transport of PAH is a process of energy dissipation. However, this conclusion still needs further study. CONCLUSION: The effect of GBE on the accumulation of PAH in renal lamina can counteract the reduction of PAH accumulation in rats with acute renal failure.
