目的:研究炎性因子C反应蛋白(CRP)和白细胞介素(IL)-6在不同年龄组大鼠主动脉组织中的表达差异,及其可能在血管衰老机制中的作用。方法:清洁级雄性SD大鼠24只,其中12只青年大鼠(青年组,2月龄)和12只老年大鼠(老年组,20月龄),经适应性喂养1周后麻醉采静脉血待测,处死后取胸主动脉组织,石蜡包埋切片HE染色后光镜下观察血管组织形态改变,Western blot检测主动脉p16和p21蛋白水平,化学发光酶免疫分析法和酶联免疫吸附试验(ELISA)分别测定大鼠血清CRP和IL-6的含量。结果:老年组大鼠主动脉组织HE染色血管中膜层增厚、血管平滑肌细胞排列紊乱,Western blot检测p16和p21蛋白表达阳性;青年组大鼠主动脉组织HE染色血管中膜层正常、血管平滑肌细胞排列整齐,p16和p21蛋白表达阴性。与青年组大鼠相比,老年组血清CRP和IL-6含量均明显升高(均P<0.05)。结论:炎症可能是导致血管衰老的原因之一。
Objective: To study the difference of expression of inflammatory factor C-reactive protein (CRP) and interleukin-6 (IL-6) in aortic tissues of different age groups and its possible role in the mechanism of vascular aging. Methods: Thirty-four male Sprague-Dawley (SD) rats were randomly divided into four groups: 12 young rats (aged 2 months) and 12 aged rats (aged 20 months old) Blood was taken and sacrificed and the thoracic aorta was taken. The paraffin embedded sections were stained with HE to observe the morphological changes of vascular tissue. Western blot was used to detect the protein levels of p16 and p21 in the aorta, chemiluminescence enzyme immunoassay and enzyme-linked immunosorbent assay The levels of CRP and IL-6 in serum were determined by ELISA. Results: HE staining in the aorta of aged rats showed thickened vascular layers and vascular smooth muscle cells arranged in a disorderly manner, and positive expressions of p16 and p21 proteins were detected by Western blot. The aortic tissue of young rats was stained with HE and the blood vessels Smooth muscle cells arranged neatly, p16 and p21 protein negative. Compared with the rats in the youth group, the levels of serum CRP and IL-6 in the elderly group were significantly increased (all P <0.05). Conclusion: Inflammation may be one of the causes of vascular aging.
