目的观察银杏叶提取物预处理对兔心肌缺血再灌注前后 PMN(中性粒细胞)的活性变化过程。了解银杏叶提取物保护心肌是否与影响中性粒细胞活化作用有关、方法取30只日本大耳白兔随机均分为3组,假手术对照组、缺血再灌注组和银杏叶提取物预处理用药组。用药方法;用自制的银杏叶提取物注射剂(总黄酮含量5mg/ml)5ml静滴给药。分别于术前和冠脉结扎缺血1h后采血用布氏显微镜对比观察白细胞活化状态的变化。检测白细胞表面粘附分子 CD;,/CD_18表达度、血浆NO(一氧化氮)、 ET(内皮素)、MDA(丙二醛)、CTnl(肌钙蛋白1),电镜下心肌组织起微结构的变化。结果在布氏显微镜下白细胞活化状态的变化是;冠脉结扎前活化的中性粒细胞(PMN)不超过5%,结扎1h后活化的白细胞达于40%~60%,而GBE预处理组结扎1h后活化的白细胞不超过18%。表面粘附分子 CD11/CD_18表达率的变化趋势与布氏显微镜下白细胞活化状态的变化相一致。心肌缺血再灌注后血浆MDA、CTnl、ET含量明显升高,血浆NO明显减少。心肌细胞线粒体肿胀、嵴减少,内皮细胞肿胀,血管腔内充满红细胞、白细胞。GBE预处理组血浆MDA、CTnl、ET含量明显低于?
Objective To observe the changes of activity of PMN (neutrophil) before and after myocardial ischemia reperfusion in rabbits by pretreatment with Ginkgo biloba extract.
