目的研究RA对宫颈癌Hela细胞中Cx26和Cx43表达的影响,探讨RA对Hela细胞增殖的影响及其机制。方法培养Hela细胞,用计数法和MTT比色法观察并比较不同浓度RA对细胞增殖的影响情况,用免疫细胞化学方法检测Hela细胞中Cx26和Cx43蛋白的表达。结果①RA抑制Hela细胞增殖,抑制率与RA浓度相关。②Cx26和Cx43蛋白定位于Hela细胞的胞膜和胞质,RA组Cx26和Cx43蛋白的表达明显高于对照组(P<0.05)。结论 RA可抑制Hela细胞增殖,其机制可能与Cx26和Cx43的表达上调有关。
Objective To investigate the effect of RA on the expression of Cx26 and Cx43 in cervical cancer Hela cells and explore the effect of RA on the proliferation of Hela cells and its mechanism. Methods Hela cells were cultured, the effects of different concentrations of RA on cell proliferation were observed by counting method and MTT colorimetric method, and the expression of Cx26 and Cx43 protein in Hela cells was detected by immunocytochemistry. Results ①RA inhibited the proliferation of Hela cells. The inhibition rate was related to the concentration of RA. The expression of Cx26 and Cx43 protein in the membrane and cytoplasm of Hela cells was significantly higher than that in the control group (P <0.05). Conclusion RA can inhibit the proliferation of Hela cells, which may be related to the up-regulation of Cx26 and Cx43.
