目的研究引起四川省2009年手足口病(Hand,Foot and Mouth Disease,HFMD)的肠道病毒71型(Enterovirus Type71,EV71)的基因特征。方法对四川省2009年HFMD病例中分离到的13株EV71,进行VP1编码区逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)扩增和核苷酸序列测定,构建遗传进化树进行基因型别分析。结果 13株分离株VP1编码区全长均为891个核苷酸,编码297个氨基酸,序列比较证实为EV71。进化分析表明,四川省EV71分离株属于C4a亚型。结论四川省2009年HFMD的病原为EV71,和中国大陆其他地区2004年以来的EV71分离株同属C4a亚型,在遗传进化上非常相近。
Objective To investigate the genetic characteristics of Enterovirus Type 71 (EV71) that caused Hand, Foot and Mouth Disease (HFMD) in 2009 in Sichuan Province. Methods Thirteen EV71 isolates isolated from HFMD cases in 2009 in Sichuan Province were amplified by RT-PCR and sequenced by RT-PCR. Phylogenetic tree analysis of genotypes. Results The full-length coding region of VP1 of the 13 isolates was 891 nucleotides, encoding 297 amino acids. The sequence comparison confirmed that EV71 was EV71. Evolutionary analysis showed that EV71 isolates in Sichuan Province belonged to C4a subtype. Conclusion The pathogen of HFMD in Sichuan province was EV71 in 2009, and the EV71 isolates in other mainland China regions since 2004 belonged to C4a subtypes, which were very similar in genetic evolution.
