阿司匹林作为抗血小板的一线药物,被广泛用于缺血性脑卒中患者的二级预防。临床试验显示,5%~65%的脑卒中患者对阿司匹林治疗反应低,为阿司匹林非敏感患者。导致此现象发生存在许多潜在因素,但目前国内外研究者研究最多、影响最大的依然是遗传因素。本文分别从阿司匹林代谢通路、环氧化酶依赖性通路及非环氧化酶依赖性通路的相关受体的基因多态性与阿司匹林非敏感现象的相关性进行综述。
As first-line anti-platelet drugs, aspirin is widely used in secondary prevention of ischemic stroke. Clinical trials show that 5% to 65% of stroke patients respond less to aspirin therapy than aspirin-sensitive patients. Lead to this phenomenon there are many potential factors, but at home and abroad the most researched, the most influential is still a genetic factor. This review summarizes the association between aspirin metabolism and aspirin insensitivity, as well as the genetic polymorphism of the related receptors in the metabolic pathway, the cyclooxygenase-dependent pathway, and the non-cyclooxygenase-dependent pathway in aspirin.
