[目的]应用不同浓度结核杆菌构建兔脊柱结核模型,探索在造模中较适合的菌量。[方法]42只新西兰兔随机分为5组,实验组3组共30只,每组10只;对照组10只;空白组2只。实验组及对照组动物经弗氏完全佐剂致敏后,在L_6上终板下方钻孔,塞入明胶海绵。实验组分别注入0.1 ml浓度为10~6CFU/ml,10~7CFU/ml,10~8CFU/ml H37Rv标准菌;对照组注入0.9%的生理盐水0.1 ml;空白组不做任何处理。[结果]术后12周兔存活率:注射10~6 CFU/ml菌量兔100%、10~7CFU/ml菌量80%、10~8CFU/ml菌量30%、对照组100%、空白组100%。术后8周X线片阳性率分别为33.3%,62.5%,66.7%;CT及三维CT阳性率分别为44.4%,75%,100%。术后12周X线片阳性率分别为44.4%,75%,100%;CT及三维CT阳性率分别为44.4%,100%,100%。术后12周解剖见17只兔椎体发生骨质破坏,部分有干酪样坏死物质形成,未见有腰大肌脓肿形成;病理学检查:片状分布的干酪样坏死物质,骨小梁排列紊乱及轮廓不清,周围见上皮样细胞及多核巨细胞包绕,局部有部分死骨碎片形成,周围见大量炎细胞浸润;实验组细菌培养阳性率分别为22.2%,75%,66.7%。对照组及空白组未出现上述结核影像学及病理改变。[结论]不同浓度H37Rv标准菌构建兔脊柱结核模型存在较大差异,10~6CFU/ml成功率较低;10~8CFU/ml死亡率及并发症较高;10~7CFU/ml造模成功率高且死亡率较低,较适合兔脊柱结核模型的构建。
[Objective] To construct rabbit spinal tuberculosis model with different concentrations of Mycobacterium tuberculosis and to explore the more suitable bacteria in the model. [Method] Forty-two New Zealand white rabbits were randomly divided into 5 groups. There were 30 rabbits in each group, with 10 in each group. There were 10 rabbits in the control group and 2 rabbits in the blank group. Animals in the experimental group and control group were sensitized with Freund’s complete adjuvant, then drilled under the end plate of L_6 and stuffed with gelatin sponge. The experimental group were injected with 0.1 ml of H37Rv standard bacteria of 10 ~ 6CFU / ml, 10 ~ 7CFU / ml and 10 ~ 8CFU / ml respectively; the control group was injected 0.9% saline 0.1ml; [Results] The survival rate of rabbits after 12 weeks of operation was 100%, 10 ~ 7CFU / ml, 80%, 10 ~ 8CFU / ml, 10% Group 100%. The positive rates of X-ray at postoperative 8 weeks were 33.3%, 62.5% and 66.7% respectively. The positive rates of CT and three-dimensional CT were 44.4%, 75% and 100% respectively. The control group and the blank group did not appear the tuberculosis imaging and pathological changes. [Conclusion] There are great differences in the model of spinal tuberculosis in rabbits constructed with different concentrations of H37Rv standard bacteria, the success rate of 10 ~ 6CFU / ml is lower; the mortality and complications of 10 ~ 8CFU / ml are higher; the successful rate of 10 ~ 7CFU / ml High and low mortality, more suitable for the construction of rabbit spinal tuberculosis model.
