目的:运用随机扩增多态性技术探索4种紫珠属药用植物的遗传多态性,为其种质鉴定提供依据。方法:采用试剂盒提取法提取裸花紫珠、广东紫珠、大叶紫珠与杜虹花4种新鲜植物叶中的总DNA,采用经过筛选的20条随机引物进行PCR扩增。结果:试剂盒可有效获取4种紫珠植物的总DNA,扩增得到条带共152条,多态性条带137条,多态率达到88.2%;聚类分析结果表明,4种紫珠供试材料共分为3类。结论:4种紫珠RAPD扩增后的聚类分析结果与植物形态学观察分类结果相一致,所得RAPD标记可用于紫珠属药用植物的多态性研究,为开发遗传鉴定的分子标记奠定基础。
OBJECTIVE: To explore the genetic polymorphisms of four species of Medicinal plants of Zizyphus by random amplified polymorphic DNA (RFLP) technique and to provide basis for their germplasm identification. Methods: Total DNA was extracted from the leaves of four fresh plants, namely Radix unguiculatus, Radix et Rhizoma, Radix et Rhizoma Rhei and Du Honghua by kit extraction method. Twenty randomly selected primers were used for PCR amplification. Results: The kit could effectively obtain the total DNA of four species of Vitex. The total number of bands amplified was 152 and the number of polymorphic bands was 137, with a polymorphism rate of 88.2%. Cluster analysis showed that four kinds of Violet Beads Test materials are divided into three categories. CONCLUSION: The results of cluster analysis after RAPD amplification of four kinds of purple pearls are consistent with the results of plant morphological observation and classification. The obtained RAPD markers can be used to study the polymorphism of medicinal plants from the genus Umerosa and lay a foundation for the development of molecular markers for genetic identification basis.
