目的本研究探讨复方君子汤(FFJZ)对抑制白血病K562细胞生长增殖和诱导凋亡的作用机制。方法用不同浓度复方君子汤作用于K562细胞,应用MTT法观察复方君子汤对K562细胞生长增殖的影响,Annexin V/PI染色和流式细胞仪(FCM)检测细胞凋亡,Western blot检测Caspase-8蛋白及Fas蛋白的表达。结果在一定浓度的复方君子汤作用后的K562细胞的增殖被抑制,随着浓度的增加及使用时间的延长,K562细胞的增殖抑制率及凋亡比例明显增加;同时,Caspase-8蛋白及Fas蛋白的表达明显上升。结论一定浓度的复方君子汤可抑制K562细胞增殖并诱导其凋亡,其机制可能与上调Caspase-8蛋白及Fas蛋白的表达有关。
Objective To investigate the mechanism of FuJijunzi Decoction (FFJZ) on inhibiting the growth, proliferation and apoptosis of leukemia K562 cells. Methods K562 cells were treated with different concentrations of compound Gentleman Decoction. MTT assay was used to observe the effect of Fufang Junzi Decoction on the proliferation and proliferation of K562 cells. Annexin V / PI staining and flow cytometry (FCM) were used to detect the apoptosis of K562 cells. Caspase- 8 protein and Fas protein expression. Results The proliferation of K562 cells treated with compound Junzi Tang was inhibited at a certain concentration. With the increase of concentration and the prolonged use of time, the proliferation inhibition rate and apoptosis ratio of K562 cells increased obviously. Meanwhile, the expression of Caspase-8 protein and Fas Protein expression increased significantly. Conclusion Fufangjunzitang can inhibit the proliferation and induce the apoptosis of K562 cells. The mechanism may be related to the up-regulation of the expression of Caspase-8 and Fas protein.
