[目的]探讨熊去氧胆酸(UDCA)对甲胎蛋白异质体(AFP-L3)及大鼠肝组织与血液中谷氨酰转移酶mRNA-H(GGTmRNA-H)亚型表达的影响,进一步研究UDCA对肝癌发生的影响。[方法]Wister大鼠92只随机分成5组:模型组(24只)、干预A组(24只)、干预B组(24只)、正常对照组(10只)、UDCA对照组(10只)。对模型组、干预A组、干预B组以二乙基亚硝铵腹腔注射诱导肝癌形成后,干预A组给予含2.5g/L的UDCA饮用水,干预B组给予含5g/L UDCA的饮用水;正常对照组给予0.9%氯化钠溶液腹腔注射,UDCA对照组给予含5g/L UDCA的饮用水。通过ELISA法检测各组大鼠血清中AFP-L3的水平,采用实时定量PT-PCR检测GGTmRNAH亚型在各组大鼠血液及肝组织的表达情况。[结果]1模型组、干预A组、干预B组AFP-L3绝对值明显高于正常对照组、UDCA对照组(P<0.05)。干预A组、干预B组AFP-L3水平均低于模型组(P<0.05)。干预A组、干预B组间比较差异无统计学意义(P>0.05)。2干预A组、干预B组中GGTmRNA-H亚型在癌组织及癌旁组织的表达量均较模型组明显下降(P<0.05);干预A组、干预B组血液中的GGTmRNA-H亚型也较模型组下降(P<0.05);但干预A组、干预B组间比较差异无统计学意义(P>0.05)。[结论]1UDCA能降低大鼠肝癌发生过程中AFP-L3的水平,提示UDCA对肝癌的发生有一定的预防作用;2UDCA能降低肝癌大鼠肝癌组织及血液中GGTmRNA-H亚型的表达,二者间接反映UDCA能降低肝癌的发生,对肝癌的发生有一定预防作用。
[Objective] To investigate the effect of ursodeoxycholic acid (UDCA) on the expression of AFP-L3 and GGT mRNA-H (GGT mRNA-H) subtypes in rat liver tissue and blood, Further study of UDCA on the occurrence of liver cancer. [Methods] 92 Wister rats were randomly divided into 5 groups: model group (24 rats), intervention group A (24 rats), intervention group B (24 rats), normal control group (10 rats), UDCA control group ). After model group, intervention group A and intervention group B, intraperitoneal injection of diethylnitrosamine (NH4Na) was used to induce the formation of hepatocellular carcinoma (HCC), group A received 2.5g / L UDCA drinking water and group B received 5g / L UDCA Water; normal control group was given 0.9% sodium chloride solution by intraperitoneal injection, UDCA control group was given 5g / L UDCA drinking water. The serum level of AFP-L3 in each group was detected by ELISA. The expression of GGT mRNA subtype in blood and liver tissues of each group was detected by real-time quantitative PCR. [Results] 1 The absolute value of AFP-L3 in model group, intervention group A and intervention group B was significantly higher than that in normal control group and UDCA control group (P <0.05). The levels of AFP-L3 in intervention group A and intervention group B were lower than those in model group (P <0.05). There was no significant difference between intervention group A and intervention group B (P> 0.05). The expression of GGT mRNA-H subtype in intervention group A and intervention group B was significantly lower than that in model group (P <0.05), while GGT mRNA-H subgroup in intervention group A and intervention group B (P <0.05). However, there was no significant difference between intervention group A and intervention group B (P> 0.05). [Conclusion] 1UDCA can reduce the level of AFP-L3 in rat hepatocarcinogenesis, suggesting that UDCA can prevent hepatocarcinogenesis. 2UDCA can reduce the expression of GGTmRNA-H subtype in hepatoma tissues and blood of rats with liver cancer. Indirectly reflect the UDCA can reduce the incidence of liver cancer, the occurrence of liver cancer have a preventive effect.
