目的:观察血红素加氧酶(HO)-1在大承气汤(DD)改善脂多糖(LPS)所致小鼠急性肺损伤(ALI)中的作用。方法:将75只雄性昆明种小鼠随机分为对照组、LPS组(经气管内滴注LPS复制ALI模型)、DD+LPS组、DD+LPS+ZnPP(锌原卟啉,HO-1特异性阻断剂)组和DD组。各组小鼠鼠于给药后6 h处死。测定肺系数;光镜观察肺组织形态学改变;检测支气管肺泡灌洗液(BALF)的中性粒细胞(PMN)数目和蛋白含量的变化;采用RT-PCR和Westernblot方法检测肺组织中HO-1 mRNA和蛋白表达的变化。结果:气管内滴注LPS可引起小鼠肺组织明显的形态学改变;BALF中PMN数目和蛋白含量均增加。肺组织中HO-1 mRNA和蛋白表达均增加。预先给予DD再气管内滴注LPS,肺组织损伤减轻,BALF中PMN数目和蛋白含量均减少,而肺组织中HO-1 mRNA和蛋白表达与LPS组相比均增加。HO-1抑制剂ZnPP可抑制DD改善肺损伤的作用。结论:DD改善LPS所致小鼠ALI的作用与其可上调HO-1 mRNA和蛋白表达有关。
Objective: To observe the effect of heme oxygenase (HO) -1 in improving the acute lung injury (ALI) induced by lipopolysaccharide (LPS) in Dachengqi Decoction (DD). Methods: Seventy-five male Kunming mice were randomly divided into control group, LPS group (LPS ALI model), DD + LPS group, DD + LPS + ZnPP Sex blockers) group and DD group. Mice in each group were sacrificed at 6 hours after their administration. The number of pulmonary neutrophils (PMN) and the content of protein in bronchoalveolar lavage fluid (BALF) were detected by RT-PCR and Western blot. The levels of HO- 1 mRNA and protein expression changes. Results: Intratracheal instillation of LPS caused obvious morphological changes in lung tissue of mice; the number of PMN and protein content in BALF increased. HO-1 mRNA and protein expression were increased in lung tissue. Pretreatment with DD followed by intratracheal instillation of LPS led to a decrease in lung tissue damage, a decrease in PMN number and protein content in BALF, and a decrease in HO-1 mRNA and protein expression in lung tissue compared with LPS group. The HO-1 inhibitor ZnPP inhibits the effects of DD on lung injury. CONCLUSION: DD can improve ALI induced by LPS in mice and increase the expression of HO-1 mRNA and protein.
