肿瘤干细胞标记物CD44~+CD24~(low/-)与MMP-9在乳腺癌组织中的表达及其意义

目的研究肿瘤干细胞标记物CD44+CD24low/-以及基质金属蛋白酶-9(MMP-9)的表达与肿瘤临床病理参数之间的关系。方法采用免疫组织化学方法检测109例乳腺癌组织中的CD44、CD24及MMP-9的表达并对结果进行分析。结果(1)109例乳腺癌组织中CD44、CD24及MMP-9的表达率分别是47.7%、50.5%及40.4%,其中CD44的阳性表达率与乳腺癌病理分级密切相关(P<0.05);(2)109例乳腺癌组织中,CD44+CD24low/-肿瘤干细胞标记阳性例数为32例,随乳腺癌病理分级的升高,CD44+CD24low/-肿瘤干细胞比率也明显增加(P<0.05);(3)32例CD44+CD24low/-肿瘤干细胞标记阳性乳腺癌组织中,MMP-9蛋白的表达与淋巴结转移成负相关(P<0.05)。结论高度恶性乳腺癌组织中富于肿瘤干细胞,乳腺癌早期CD44+CD24low/-TSC中MMP-9蛋白表达增加可能对肿瘤干细胞(TSC)生长及浸润中起着一定作用。 Objective To study the relationship between the expression of tumor stem cell markers CD44 + CD24low / - and matrix metalloproteinase-9 (MMP-9) and clinicopathological parameters of tumors. Methods Immunohistochemistry was used to detect the expression of CD44, CD24 and MMP-9 in 109 cases of breast cancer and the results were analyzed. Results (1) The positive rates of CD44, CD24 and MMP-9 in 109 breast cancer tissues were 47.7%, 50.5% and 40.4%, respectively. The positive rate of CD44 was closely related to the pathological grade of breast cancer (P <0.05). (2) Of the 109 cases of breast cancer, the positive number of CD44 + CD24low / - tumor stem cells was 32 cases. With the increase of the pathological grade of breast cancer, the percentage of CD44 + CD24low / - tumor stem cells was also significantly increased (P <0.05) ; (3) In 32 cases of CD44 + CD24low / - cancer stem cell marker positive breast cancer, the expression of MMP-9 protein was negatively correlated with lymph node metastasis (P <0.05). Conclusions The increased expression of MMP-9 in early stage CD44 + CD24low / -TSC in breast cancer may play a role in the growth and infiltration of tumor stem cells (TSC) in highly malignant breast cancer.
