目的观察SIV感染CEM×174细胞后CTLA-4、FOXP3、IDOmRNA表达水平的动态变化。方法以SIVmac251感染接种CEM×174细胞,收集感染后12h~216h共8个时间点及正常未感染细胞,提取细胞RNA,进行RNA逆转录荧光定量PCR检测。结果随着SIVmRNA的升高,CTLA-4、FOXP3、IDO三者mRNA均显著升高;SIVmRNA、CTLA-4mRNA到观察结束时(216h)达到最高值,而FOXP3、IDO则是在168h时mRNA达到最高值,在216h时有所回落。结论被SIV感染的细胞中部分促进免疫激活和抑制免疫的免疫调节因子表达均显著升高,提示免疫紊乱是AIDS的一个重要发病机制。
Objective To observe the dynamic changes of CTLA-4, FOXP3 and IDO mRNA expression in SIV-infected CEM × 174 cells. Methods CEM × 174 cells were inoculated with SIVmac251, and 8 non-infected cells were collected at 12h-216h after infection. Cell RNA was extracted and detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The mRNAs of CTLA-4, FOXP3 and IDO were significantly increased with the increase of SIV mRNA. The mRNA of SIV mRNA and CTLA-4 reached the peak value at the end of observation (216h), while FOXP3 and IDO reached the peak at 168h The highest value, at 216h, has come down. Conclusions The expression of some immune-activating and immune-suppressing immune regulators in SIV-infected cells were significantly increased, suggesting that immune disorder is an important pathogenesis of AIDS.
