目的 :观察槲皮素 (Que)、异鼠李素 (Iso)对Cu2 + 介导的低密度脂蛋白 (LDL)氧化修饰的影响。方法 :采用一次性密度梯度离心法分离正常人血浆脂蛋白 ,用Cu2 + 进行体外氧化修饰 ,温育前加不同浓度的Que及Iso。检测脂蛋白中脂质过氧化物 (LPO)、维生素E(VitE)含量及超氧化物歧化酶 (SOD)活性。结果 :在Cu2 + 与LDL温育前加入Que、Iso可使LDL中LPO生成减少 ,明显延缓VitE含量的减少 ,显著提高SOD活性 ,并呈现一定的浓度效应。结论 :结果显示Que和Iso均对Cu2 + 诱导的LDL氧化修饰具有明显的抑制作用 ,但Que的作用效果强于Iso。
Objective: To observe the effect of quercetin (Que) and isorhamnetin (Iso) on Cu2 + -mediated oxidation of low density lipoprotein (LDL). Methods: The plasma lipoproteins of normal people were isolated by one-time density gradient centrifugation, and oxidized by Cu2 + in vitro. Different concentrations of Que and Iso were added before incubation. The contents of lipid peroxide (LPO), vitamin E (VitE) and superoxide dismutase (SOD) in the lipoproteins were determined. Results: The addition of Que and Iso before incubation with Cu2 + and LDL decreased the production of LPO in LDL, significantly delayed the decrease of VitE content, significantly increased the activity of SOD and showed a certain concentration effect. CONCLUSION: Both Que and Iso showed significant inhibitory effects on Cu2 + -induced LDL oxidation, but the effect of Que was stronger than that of Iso.
