目的探讨蒸制轮叶党参(steamed codonopsis lanceolata,SCL)对顺铂(Cisplatin)致小鼠肾损伤的保护作用及机制。方法雄性ICR小鼠一次性腹腔注射20 mg/kg顺铂诱发急性肾损伤模型,观察SCL给药后的改善作用。相关试剂盒测定小鼠血清中肌酐(CRE)、尿素氮(BUN)、炎症因子(TNF-α)、白介素1β(IL-1β)和肾组织中丙二醛(MDA)的含量。苏木素-伊红(HE)和Hoechst 33258染色观察肾组织病理学改变。免疫组化法检测肾组织中相关凋亡蛋白的表达。结果与模型组比较,SCL均能不同程度的降低血清中CRE、BUN、TNF-α、IL-1β及组织中MDA的水平(P<0.05),改善顺铂所致的炎症反应,同时SCL能够下调肾组织中Bax,上调Bcl-2的蛋白表达水平。结论 SCL对顺铂致肾损伤具有保护作用,其主要部分通过减少炎症反应,抑制脂质过氧化及抗细胞凋亡而实现的。
Objective To investigate the protective effect of steamed codonopsis lanceolata (SCL) on renal injury induced by Cisplatin and its mechanism. Methods The acute renal injury model was induced by intraperitoneal injection of 20 mg / kg cisplatin in male ICR mice and the improvement after SCL administration was observed. The related kit was used to determine the levels of serum creatinine (CRE), blood urea nitrogen (BUN), inflammatory cytokines (TNF-α), interleukin-1β (IL-1β) and malondialdehyde (MDA) Hematoxylin-Eosin (HE) and Hoechst 33258 staining were used to observe the histopathological changes of renal tissue. Immunohistochemistry was used to detect the expression of related apoptosis proteins in renal tissues. Results Compared with the model group, SCL decreased the levels of CRE, BUN, TNF-α, IL-1β and MDA in the tissue (P <0.05) to some extent and improved the cisplatin-induced inflammatory response Down-regulate Bax in renal tissue, and up-regulate the protein expression of Bcl-2. CONCLUSIONS: SCL has a protective effect on Cisplatin-induced renal injury. The main part of SCL can reduce inflammation, inhibit lipid peroxidation and prevent apoptosis.
