目的:研究羧甲基壳聚糖(CMC)对雌二醇凝胶体外透皮吸收的影响。方法:合成CMC,用电位滴定法确定其取代度。以含2%CMC的雌二醇凝胶为试验组,含2%氮酮的雌二醇凝胶为阳性对照组,无任何促透剂的雌二醇凝胶为阴性对照组,通过体外透皮扩散实验得到各组累积透过量,并求出稳态透皮速率。结果:合成得到的CMC取代度为0.52。试验组的累积透过量和稳态透皮速率显著大于阴性对照组(P<0.05),但与阳性对照组相比,差异无统计学意义(P>0.05)。结论:2%CMC与2%氮酮的促透效果相似,值得进一步研究。
Objective: To study the effect of carboxymethyl chitosan (CMC) on transdermal absorption of estradiol gel in vitro. Methods: CMC was synthesized and its degree of substitution was determined by potentiometric titration. Estradiol gel with 2% CMC was used as the test group, estradiol gel with 2% Azone was used as the positive control group, estradiol gel without any enhancer was used as the negative control group, The skin penetration experiment obtained the cumulative permeation of each group, and found the steady-state transdermal rate. Results: The degree of substitution of CMC was 0.52. The cumulative permeation rate and steady-state transdermal rate of the experimental group were significantly higher than those of the negative control group (P <0.05), but there was no significant difference between the experimental group and the positive control group (P> 0.05). Conclusion: 2% CMC and 2% Azone have the same promoting effect and deserve further study.
