目的探讨模拟失重30 d及再超重对猕猴腮腺组织中热休克蛋白HSP70表达的影响。方法选用雄性猕猴23只,随机分为对照组(A组),失重(B组),超重+13 Gx/230s(C组),失重后再超重(D组,依据不同的+Gx强度和时间分为4个亚组:分别是+11 Gx/270s D1组、+13 Gx/230s D2组、+15 Gx/200s D3组、+13 Gx/230s后恢复9 d D4组)。通过HE染色观察腮腺组织形态学的变化,通过免疫组织化学染色及Western blot技术检测HSP70在腮腺中的表达。采用SPSS17.0对数据进行统计分析。结果对照组腮腺结构基本正常,实验组腮腺组织中可见不同程度的局灶性淋巴细胞浸润、腺泡细胞萎缩,导管扩张,血管扩张充血;对照组腮腺组织HSP70表达阴性,实验组腮腺组织HSP70表达阳性,随着+Gx强度的加大而表达增强的趋势。实验组(除D4组外)HSP70在蛋白水平的表达显著高于对照组(P<0.05)。结论模拟失重、超重环境可造成猕猴腮腺出现轻度病理变化,腮腺组织中HSP70表达显著增强。
Objective To investigate the effects of 30-day simulated weightlessness and re-overweight on heat shock protein HSP70 expression in parotid glands of rhesus monkeys. Methods Twenty-three male cynomolgus macaques were randomly divided into control group (group A), weightless group (B), overweight +13 Gx / 230s group C, and then overweight after weightlessness (group D according to different + Gx intensity and time Divided into 4 subgroups: +11 Gx / 270s D1 group, +13 Gx / 230s D2 group, +15 Gx / 200s D3 group, and 9 d D4 group after +13 Gx / 230s. The changes of parotid gland morphology were observed by HE staining. The expression of HSP70 in the parotid gland was detected by immunohistochemical staining and Western blot. Data were analyzed by SPSS17.0. Results The structure of parotid gland in the control group was basically normal. The parotid gland in the experimental group showed varying degrees of focal lymphocyte infiltration, acinar cell atrophy, catheter dilatation and vasodilation and congestion. The expression of HSP70 in the parotid gland of the control group was negative, Positive, with increasing intensity of + Gx expression increased trend. The expression of HSP70 in the experimental group (except the D4 group) at the protein level was significantly higher than that of the control group (P <0.05). Conclusion Simulated weightlessness and overweight environment can cause mild pathological changes in the parotid gland of the rhesus monkey, and the expression of HSP70 in the parotid gland is significantly increased.
