本文比较分析了二糖蔗糖、麦芽糖、乳糖和单糖葡萄糖、果糖、半乳糖共6种碳源;0.8%琼脂,0.2%Gelrite和液体三种凝固剂处理对棉花胚珠Bt培养基pH值的影响。试验结果表明,培养基分别经高温高压灭菌和贮存6周后,其pH值均有不同程度的降低。如果综合考虑碳源和凝固剂对培养基pH值的影响,那么在碳源中二糖就应采用蔗糖,在单糖中就应采用葡萄糖;在凝固剂中以采用0.8%琼脂较好。碳源和凝固剂通过对培养基pH的影响而会最终影响棉花胚珠离体纤维的发育。
In this paper, six carbon sources such as sucrose, maltose, lactose and monosaccharides glucose, fructose and galactose were analyzed. Three kinds of coagulants including 0.8% agar, 0.2% Gelrite and liquid were used to treat cotton ovule Bt medium Effect of pH. The results showed that after the medium was sterilized by autoclave and stored for 6 weeks respectively, the pH value of the medium decreased to some extent. If we consider the impact of carbon source and coagulant on the pH of the medium, then the disaccharides should be used in the carbon source of sucrose, glucose should be used in the monosaccharide; coagulant to use 0.8% agar is better . The effect of carbon source and coagulant on the pH of the medium will ultimately affect the development of the fiber in vitro in cotton ovules.
