目的:探讨趋化因子CCL19在人头颈鳞癌细胞活性中的作用。方法:应用噻唑蓝比色(MTT)法检测不同浓度顺铂对人头颈鳞癌原发细胞系(PCI-4A)及淋巴结转移细胞系(PCI-4B)的生长抑制作用,观察不同浓度的CCL19对顺铂作用的影响,应用透射电镜技术观察4B细胞形态学变化,流式细胞仪(FCM)检测CCL19对顺铂诱导的4B细胞周期及凋亡率的影响。使用SPSS11.0软件包进行统计学分析。结果:顺铂对4B细胞的生长抑制率高于4A细胞,CCL19可以显著降低顺铂对4B细胞的生长抑制(P<0.05),但对4A细胞无明显作用(P>0.05),CCL19对顺铂作用的4B细胞的影响是通过形态学上向正常状态转化、减少细胞在G1期的停滞、减少细胞的凋亡率而产生的。结论:顺铂在人头颈鳞癌淋巴结转移细胞(PCI-4B)中的作用强于原发细胞(PCI-4A),CCL19可以显著降低顺铂在PCI-4B细胞中的作用,提高细胞的活性,这为探讨临床头颈鳞癌的化疗耐药机制提供了一个新的思路。
Objective: To investigate the role of chemokine CCL19 in the activity of human head and neck squamous cell carcinoma. Methods: The inhibitory effects of different concentrations of cisplatin on primary head and neck squamous cell carcinoma (PCI-4A) and lymph node metastasis cell line (PCI-4B) were detected by MTT assay. The effects of different concentrations of CCL19 On the effect of cisplatin, the morphological changes of 4B cells were observed by transmission electron microscopy. The effect of CCL19 on cisplatin-induced 4B cell cycle and apoptosis rate was examined by flow cytometry (FCM). SPSS11.0 software package for statistical analysis. Results: The inhibitory rate of cisplatin on 4B cells was higher than that of 4A cells. CCL19 could significantly inhibit the growth of 4B cells (P <0.05), but had no effect on 4A cells (P> 0.05) The effect of platinum-induced 4B cells is generated by morphological changes to normal, reducing cell arrest in the G1 phase and decreasing the rate of apoptosis of cells. CONCLUSIONS: Cisplatin is more effective than PCI-4B in lymph node metastasis of human head and neck squamous cell carcinoma (PCI-4B). CCL19 can significantly reduce the effect of cisplatin in PCI-4B cells and improve cell activity , Which provides a new idea for exploring the chemical resistance mechanism of clinical head and neck squamous cell carcinoma.
