成年雄性Wistar大鼠54只,随机分为实验性胃溃疡组、盐水对照组和空白对照组。前两组又各分为术后4天、6天、10天及28天组。取垂体,用Bouin液固定,制成石蜡切片,用Sternberger PAP法显示垂体远侧部的ACTH细胞。在光镜下,用点分析法测量各组ACTH细胞的面数密度和体积密度,以观察细胞数量和体积的变化。用图像分析仪对6天溃疡组、6天盐水组和空白对照组动物的ACTH细胞进行光密度测量,以观察细胞激素含量的变化。结果显示,实验性胃溃疡组术后4天ACTH细胞的数量,比空白对照组明显减少;术后6天,细胞数量明显增多;术后10天,细胞数量开始下降,
Fifty-four adult male Wistar rats were randomly divided into experimental gastric ulcer group, saline control group, and blank control group. The first two groups were divided into 4 days, 6 days, 10 days and 28 days after operation. The pituitary gland was removed and fixed with Bouin solution to make a paraffin section. The ACTH cells in the distal part of the pituitary were visualized by the Sternberger PAP method. Under the light microscope, the spot number method was used to measure the surface number density and bulk density of each group of ACTH cells to observe changes in cell number and volume. The optical density of ACTH cells in the 6-day ulcer group, the 6-day saline group, and the blank control group was measured with an image analyzer to observe changes in cytokine levels. The results showed that the number of ACTH cells in the experimental gastric ulcer group was significantly lower than that in the blank control group on the 4th day after operation; the number of cells significantly increased on the 6th day after operation; the number of ACTH cells began to decrease after 10 days.
