目的 :研究碱性成纤维细胞生长因子 (bFGF)在乳腺癌细胞中的表达特征 ,寻找有助于诊断乳腺癌的生物学标记 .方法 :收集乳腺腺病、乳腺纤维腺瘤和乳腺癌等的手术切除标本各 2 2例 ,常规石蜡包埋切片 (4μm) ,分别做HE染色和用ABC法进行bFGF(1∶2 0 0 )、血管标记抗体CD34(1∶10 0 )免疫组织化学染色 ,参考Weidner和Maeda方法进行微血管密度 (MVD)测定 .结果 :在乳腺腺病和乳腺纤维腺瘤中的肌上皮细胞、导管上皮细胞、血管内皮细胞、血管壁平滑肌细胞和纤维母细胞等均表达bFGF ,其阳性产物主要位于胞浆 ,肌上皮细胞和导管上皮细胞的阳性产物部分见于胞核 ,但乳腺癌细胞的阳性产物主要位于胞膜 ,少数位于胞浆 ;胞核阴性 ,与良性增生性病变形成鲜明的对照 .2 2例乳腺癌bFGF膜阳性者 17例 (占 77 3% ) ,其MVD =90 5 6± 2 3 6 2 .肿瘤直径 4cm(10 /2 2例 )时 ,MVD增高达 93 19± 2 0 75 .乳腺腺病MVD =4 2 0 5± 12 2 6 ,乳腺纤维腺瘤MVD =4 3 14± 18 2 1,与乳腺癌相比有显著差异 (P <0 0 1) .结论 :1)bFGF免疫染色阳性产物在乳腺癌细胞中主要分布在胞膜 ,在良性增生性病变细胞中主要位于胞浆和胞核 .乳腺癌的MVD明显高于良性增生性病变 .2 )质量好的免疫染色、bFGF反应产物的分布特征 ,结合MVD有助于
OBJECTIVE: To study the expression of basic fibroblast growth factor (bFGF) in breast cancer cells and to find out the biomarkers that may be useful for the diagnosis of breast cancer.Methods: The breast adenocarcinoma, breast fibroadenoma and breast cancer were collected Twenty-two surgical specimens were removed and paraffin-embedded sections (4 μm) were used for HE staining and immunohistochemical staining of bFGF (1: 200) by ABC method and CD34 (1:10 0) The microvessel density (MVD) was measured by the method of Weidner and Maeda.Results: The myofibroblasts, ductal epithelial cells, vascular endothelial cells, vascular smooth muscle cells and fibroblasts in breast adenosis and breast fibroadenoma all expressed bFGF , The positive products mainly located in the cytoplasm, myoepithelial cells and ductal epithelial cells in the positive product part of the nucleus, but the positive product of breast cancer cells located mainly in the cell membrane, a few in the cytoplasm; nuclear negative, and benign proliferative lesions (P <0.05) .2 cases of breast cancer bFGF membrane positive in 17 cases (77 3%), the MVD = 905 6 ± 2 362. Tumor diameter 4cm (10/222 cases), MVD increased up to 93 19 ± 2 0 75. MVD was 4250 ± 1226, MVD was 4314 ± 1821, which was significantly different from that of breast cancer (P <0.01) .Conclusions: 1) bFGF immunization The positive staining products in breast cancer cells mainly distributed in the membrane, mainly in the cytoplasm and nucleus in benign proliferative cells, MVD in breast cancer was significantly higher than that in benign proliferative lesions.2) Good quality immunostaining, bFGF The distribution of reaction products, combined with MVD, helps
