本文应用气溶胶转鼓设备,观察了大肠杆菌基因工程株(MM3)和标准株8099在空气中的稳定性。实验结果表明,在温度20℃,相对湿度(RH)60%的条件下,气溶胶储存60min,MM3和8099的平均存活率比值为0.4:1,t检验,差别高度显著(P<0.01)。用含有氯霉素的LB琼脂测定MM3在空气中质粒的丢失数量,未发现质粒丢失。无论在日光总辐射、散射,还是黑暗环境,土壤中的工程株都比标准株衰亡快。
In this paper, aerosol drum equipment was used to observe the stability of Escherichia coli genetically engineered strain (MM3) and standard strain 8099 in air. The experimental results showed that the average viability ratio of MM3 and 8099 was 0.4: 1 at 60 ℃ for 60min, and the difference was highly significant (P <0 .01). The amount of MM3 plasmid lost in air was determined using LB agar containing chloramphenicol and no plasmid loss was found. Whether in the total solar radiation, scattering, or dark environment, engineering strains in the soil faster than the standard strain decline.
