目的探讨人骨肉瘤组织中DNA损伤修复基因以及TS、β-tubulinⅢ多个化疗反应相关标志物的表达谱特点,及其与临床病理的关系。方法选取50例诊断明确的骨肉瘤患者组织蜡块,运用组织芯片和免疫组化技术检测OGG1、BRCA1、RRM1、XRCC1、APE1、ERCC1以及TS、β-tubulinⅢ8种基因在骨肉瘤组织中的表达情况。结果①OGG1、BRCA1、RRM1、XRCC1、APE1、ERCC1在骨肉瘤组织中呈阳性表达,阳性率分别为96%、94%、94%、84%、84%、74%,强阳性率为64%、76%、54%、72%、48%、2%,而TS仅有6例有弱阳性表达,β-tubulinⅢ无阳性表达。②阳性表达的基因中,仅APE1表达与骨肉瘤的组织病理类型具有相关性(P<0.05)。③等级相关分析,APE1和RRM1、APE1和OGG1、OGG1和BRCA1表达呈正相关(r=0.342、0.318、0.319,P<0.05),BRCA1和ERCC1表达呈负相关(r=-0.324,P<0.05);APE1和XRCC1、RRM1和XRCC1的表达也呈正相关(r=0.406、0.677,P<0.01)。结论骨肉瘤组织中存在多种DNA损伤修复相关基因的表达,其中部分基因的表达强弱之间具有相关性,APE1的表达强弱与肺癌病理类型有关。
OBJECTIVE: To investigate the expression profiles of DNA damage repair genes and related markers of TS, β-tubulinⅢ in human osteosarcoma tissues and their relationship with clinical pathology. Methods The expression of OGG1, BRCA1, RRM1, XRCC1, APE1, ERCC1 and TS, β-tubulin Ⅲ in osteosarcoma tissues were detected by tissue microarray and immunohistochemistry in 50 cases of osteosarcoma. . Results The positive rates of OGG1, BRCA1, RRM1, XRCC1, APE1 and ERCC1 in osteosarcoma tissues were 96%, 94%, 94%, 84%, 84% and 74% 76%, 54%, 72%, 48%, 2%, while only 6 of TS had a weak positive expression and no positive expression of β-tubulinⅢ. ② Only the expression of APE1 was correlated with histopathological types of osteosarcoma (P <0.05). There was a positive correlation between the expression of APE1 and RRM1, APE1 and OGG1, OGG1 and BRCA1 (r = 0.342,0.318,0.319, P <0.05), and negative correlation between ERCC1 and BRCA1 (r = -0.324, There was also a positive correlation between the expression of APE1 and XRCC1, RRM1 and XRCC1 (r = 0.406,0.677, P <0.01). Conclusion The expression of several genes related to DNA damage repair exists in osteosarcoma tissues. The expression of some genes is correlated with each other. The expression of APE1 is related to the pathological type of lung cancer.
