Apoptosis of prostatic cells in male rats with diabetes mellitus

Objective: To assess the dynamic changes in apoptosis of the prostate in diabetic male rats. Methods: Forty rats were rendered diabetic with introperitonal injection of streptozocin (STZ) at a dose of 60 mg/kg; 1, 3, 5, 7 and 14 days later rats were killed in groups of 8 each. Another 8 normal rats served as the controls. Apoptosis were examined by TUNEL immunostaining technique and flow cytometry. Results: In the D5, D7 and D14 groups, obvious apoptotic changes can be seen in the prostatic epithelium under high power microscope and the TUNEL-positive cell rate was significantly higher as compared with the controls. However, the TUNEL labeling cell rate was significantly lower in the D3 group than in the controls. The ratio of G0/G1 cells in D3 and D5 groups was significantly lower, and in D14 group the percentage of S cells was significantly higher and the G2-M cells of this group significantly lower as compared with the controls. Conclusion: In rats, diabetes may induce changes in cell cycle and apoptosis in the prostate. The apoptotic cells were significantly reduced on D3 after STZ injection and increased significantly thereafter.
