目的研制四物汤巴布剂并探讨其体外透皮吸收。方法以水溶性高分子材料为辅料制备四物汤巴布剂,以初黏力,剥离强度为考察指标,优选巴布剂基质的最佳处方;采用Franz扩散池法,以离体小鼠皮肤作为扩散屏障,用HPLC对四物汤巴布剂中阿魏酸和芍药苷进行含量测定,考察其累积透过量。结果四物汤巴布剂的最佳处方为:卡波姆∶聚丙烯酸钠∶三乙醇胺∶高岭土∶甘油∶聚乙二醇∶三氯化铝∶药物浸膏=0.9∶0.9∶0.9∶0.6∶4∶2∶0.1∶3;采用3%的氮酮作为透皮促进剂对阿魏酸和芍药苷均有良好的透皮促进效果;阿魏酸和芍药苷的释药规律符合Higuchi方程,分别为Q=0.8433t1/2-0.2824,r=0.9603和Q=3.7426t1/2-4.1243,r=0.9788。结论该研究为四物汤外用剂型的开发及应用奠定理论基础。
Objective To develop Siwu Tang poultice and explore its transdermal absorption in vitro. Methods Water-soluble polymer materials for the preparation of Siwu Tang poultice, the initial viscosity, peel strength as the index, the best prescription for cataplasma matrix; Franz diffusion cell method to isolated mouse skin As a diffusion barrier, the content of ferulic acid and paeoniflorin in Siwu Tang was determined by HPLC, and the cumulative permeation was investigated. Results The best prescription of Siwu Tang poultice was carbomer: sodium polyacrylate: triethanolamine: kaolin: glycerol: polyethylene glycol: aluminum trichloride: drug extract = 0.9: 0.9: 0.9: 0.6: 4: 2: 0.1: 3; 3% Azone as a transdermal enhancer on the ferulic acid and paeoniflorin have a good transdermal promoting effect; ferulic acid and paeoniflorin release law in line with Higuchi equation, respectively Q = 0.8433t1 / 2-0.2824, r = 0.9603 and Q = 3.7426t1 / 2-4.1243, r = 0.9788. Conclusion This study lays a theoretical foundation for the development and application of topical formulations of Siwu Tang.
