目的探讨镉对红细胞膜损伤作用的机制。方法采用傅立叶变换红外(FTIR)漫反射光谱技术,用马来酰亚胺自旋标记(MSL)、5-氮氧自由基硬脂酸标记(5-DS)的电子自旋共振(ESR)技术进行研究。结果CdCl2对红细胞膜蛋白构象、膜脂质流动性均有影响。在0.001~0.100mmol/L剂量范围内,镉离子可使红细胞膜蛋白发生变性和不可逆聚合。结论膜蛋白的-SH基是镉作用的分子靶点;镉离子可使浅层膜脂质流动性增加。
Objective To investigate the mechanism of cadmium on erythrocyte membrane damage. Methods Fourier Transform Infrared (FTIR) Diffuse Reflectance Spectroscopy (FTIR) was used to determine the molecular weight of the product by means of Electron Spin Resonance (ESR) with 5-Nitroxide Stearic Acid Labeled (MSL) research. Results CdCl2 had an influence on membrane protein conformation and membrane lipid fluidity of erythrocytes. In the dose range of 0.001 ~ 0.100mmol / L, cadmium ions can cause denaturation and irreversible polymerization of erythrocyte membrane proteins. Conclusions -SH group of membrane proteins is the molecular target of cadmium. Cadmium ions can increase lipid fluidity of superficial membrane.
