目的建立根痛合剂中葛根素和芍药苷含量测定的高效液相色谱法(HPLC)。方法采用依利特Elite-C_(18)色谱柱(200mm×4.6 mm,5μm);流动相:乙腈-甲醇-水(15∶15∶70);流速:1.0 m L·min~(-1);检测波长:250 nm,柱温:25℃。结果葛根素在0.2676~2.676μg范围内线性关系良好(r=0.999 9),平均加样回收率为100.1%(n=6),RSD为0.8%,芍药苷在0.2024~2.024μg范围内线性关系良好(r=0.999 8),平均加样回收率为99.2%(n=6),RSD为1.1%。结论该试验所建立的方法简单可行,重复性好,可以用于控制根痛合剂的质量。
Objective To establish a HPLC method for the determination of puerarin and paeoniflorin in root pain mixture. Methods Ellite Elite-C 18 column (200 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetonitrile-methanol water (15:15:70). The flow rate was 1.0 m L · min -1 Detection wavelength: 250 nm, column temperature: 25 ℃. Results Puerarin showed good linearity (r = 0.999 9) in the range of 0.2676 ~ 2.676 μg with an average recovery of 100.1% (n = 6), RSD of 0.8% and paeoniflorin in the range of 0.2024 ~ 2.024 μg Good (r = 0.999 8), with an average recovery of 99.2% (n = 6) and a RSD of 1.1%. Conclusion The method established in this experiment is simple and feasible with good repeatability and can be used to control the quality of root pain mixture.
